I recently bought a Lasius flavus queen. She currently resides in the test tube she came in from Ant Store. I don't intend on moving her out of the tube until the first workers have hatched, but how often do I have to remove the bung to ensure she gets enough air as I'm worried that she may suffocate?!

Air does manage to permeate through a cotton wool bung, even if it is wedged tight; but you can loosen the bung for just a couple of seconds each day, as this will allow more than enough fresh air into the tube
